Array Design
- febit miRNA arrays are based on the most recent miRBase version (12.0), please refer to http://microrna.sanger.ac.uk
- febit miRNA arrays allow profiling of any miRNA in any species, including:

- capture probe design for miRNA targets uses original miRNA sequence including mismatch probes
Controls used
- endogenous small RNAs for spike-in
- artificial febit miRNA spiking controls
Sample Preparation
- samples of total RNA or RNA-enriched for the miRNAs are acceptable:
- start with as low as 250 ng total RNA (standard : 1-2µg)
- use protocols for isolation of mature miRNAs or small RNA fraction (<200bp)
- use the >200bp fraction for mRNA profiling of the same sample
- ship samples to febit on dry ice via express service
- febit performs miRNA sample labeling with an optimized Genisphere Kit
Sample QC
- incoming total RNA samples are checked for quality before labeling. Samples failing QC will result in the notification of customer for potential resubmission of sample
Hybridization
- febit employs an optimized miRNA buffer for incubation
- sensitivity is increased through active hybridization
Detection
- biotin-SAPE system allows for the highest sensitivity
- febit´s fully automated antibody amplification procedure gives increased sensitivity (factor of ~5-7)
Data Analysis
- raw data export (.xls, .txt, image files) for analysis with external software at customer´s convenience
- optional full data analysis according to customer´s requirements
Sensitivity
- optimized experimental protocols yield excellent results using as little as 250 ng of total RNA
- minimum amount of miRNA that can reliably be detected when spiked into 5 µg total RNA: approx. 500 fg
- sensitivity: 0.2 attomoles
Specificity
- high specificity has been demonstrated through the clear discrimination obtained between the highly homologous members of the let7 family
- minimal difference of miRNA expression between two samples that can be determined with statistical significance: ~30%
